Chicago would be an ~8 hour drive, but not sure what else to suggest, not a ton of options that meet the 5 hour part. Even Detroit is a 6 hour drive (4 if you cut through Canada I guess)
I don’t believe any city within a 5 hour drive of Buffalo will give you what you want. You would need to go to the Atlantic Coast, Gulf Coast, West Coast, or Chicago.
I think you should be more clear in what exactly you want out of a city. It’s hard comparing any city to Miami, never mind one being near Buffalo, and I don’t know what similarity Niagara Falls has that you like.

Cleveland, honestly. It has a small but nice skyline and plenty of good beaches—not Florida quality, but excellent Great Lakes beaches. Inexpensive and close to your parents. Big city vibe (compared to Jamestown) but not overwhelming.
